Tsui et al. (1996) locirali su gen CRIP2 na hromosomsku poziciju 14q32, primjenom fluorescentne in situ hibridizacije (FISH).[9] Analizom somatskih hibridnih ćelija i FISH-metoda, Karim et al. (1996) mapirali su gen CRIP2 na poziciji 14q32.3.[10]
CRIP2 i usko povezani CRIP1 su proteini bogati cisteinom i sadrže po dva LIM-domena. Visoko su eksprimirani tokom kardiovaskularnog razvoja i djeluju na premošćavanje faktora serumskog odgovora i GATA i stimuliraju ciljne gene glatkih mišića.[11]
Skriningom niske strogosti ljudske biblioteke cDNKtankog crijeva cDNK bogatim cisteinom (CRIP1) cDNK, gorepomenuti Karim et al (1996) izolirali su cDNK koja kodira CRIP2, koju su nazvali ESP1/CRP2. cDNK predviđa protein od 208 aminokiselina koji sadrži dva LIM-domena. CRIP2 dijeli 93% identiteta aminokiselinske sekvence sa svojim homologom kod pacova, zvanim Esp1 ili Crp2. Northern blot analiza pokazala je široko rasprostranjenu tkivnu ekspresiju CRIP2 iRNK od 1,3 kb, s najvišim nivoom ekspresije u srcu. Također otkrivena je o druga kRNK u sjemenicima, od 1,7 kb.
Tsui et al. (1996) klonirali su cDNK ljudskog srca koja kodira CRIP2 i nazvali je CRP2. CRIP2 je 38%, odnosno 35% identičan proteinima CSRP1 i CSRP3.
