Kloniranje i ekspresija
Koristeći serijsku analizu ekspresija gena (SAGE), St Croix et al. (2000) identificirali su parcijalne cDNK, koje odgovaraju za nekoliko tumorskih endotelnih markera (TEM) koji su pokazali povišenu ekspresiju tokom tumorske angiogeneze. Među genima koje su identificirali bio je PLXDC1, koji su nazvali TEM7. Korištenjem pretraživanja baze podataka i RACE-a s pet najboljih, Carson-Walter et al. (2001) izveli su sekvence cijele regiju kodiranja TEM7, koja kodira 500-aminokiselinski tip I transmembranskog proteina koji sadrži domen sličan pleksinu. Zamjenski transkript gena TEM7 označili su TEM3. TEM3 i TEM7 se razlikuju u upotrebi alternativnih mjesta poliadenilacije, ali rezultiraju istim predviđenim proteinima. Mišji ortolog TEM7 dijeli 81% aminokiselinskih identiteta s ljudskim proteinima.
Analiza hibridizacije na ljudskog karcinoma debelog crijeva od Carson-Walter et al. (2001) pokazali su da se TEM7 jasno eksprimira u endotelnim ćelijama tumorske strome, ali ne i u endotelnim ćelijama normalnog tkiva debelog crijeva. Koristeći fluorescentnu hibridizaciju in situ za ispitivanje ekspresije u raznim normalnim tkivima odraslog miša, primijetili su da Tem7 uglavnom nije moguće otkriti u tkivima ili tumorima, ali je obilno eksprimiran u mišjem mozgu. Lokalizirali su ekspresiju Tem7 u Purkinjeovim ćelijama malog mozga i nekim neuronskim ćelijama.[9][10]
