YAF2
YY1-pridruženi faktor 2 je protein koji je kod ljudi kodiran genom YAF2.[5][6] Gen YAF2 mapiran je na hromosomu 12q12, na osnovu poravnavanja sekvence YAF2 (GenBank BC037777) sa genomskom sekvencom (GRCh38).
Protein kodiran ovim genom stupa u interakciju s YY1, proteinom cinkovog prsta, koji je uključen u negativnu regulaciju gena ograničenih na mišiće. Sam ovaj genski proizvod sadrži jedan N-terminalni C2-X10-C2 cinkov prst, i za razliku od YY1, nsdreguliran je tokom miogene diferencijacije. Također olakšava proteolitsko cijepanje YY1 pomoću proteaze aktivirane kalcijem, m-kalpaina, sugerirajući mehanizam pomoću kojeg ovaj protein antagonizira negativaom učinku YY1.[6]

Kloniranje i ekspresija
Koristeći YY1 kao sondu u 2-hibridnom kvascu, Kalenik et al. (1997) klonirali YAF2 iz biblioteke cDNK mišića. Izvedeni protein od 180 aminokiselina ima izračunatu molekulsku masu od oko 20 kD. Sadrži N-terminalni motiv cinkovog prsta tipa C2-X10-C2, gdje je X bilo koja aminokiselina, i navodni signal jedarne lokalizacije. YAF2 dijeli homologiju ograničene sekvence s nekoliko nukleoporina i RAN-vezujućeg proteina-2 (RANBP2).[8]
Prema Northern blot analizi, Sawa et al. (2002) otkrili su ekspresiju YAF2 u mnogim tkivima, s najvećom ekspresijom u srcu i skeletnim mišićima. Uočili su homologiju između YAF2 i RYBP, posebno unutar njihovih N-terminalnih domena.[9]
Funkcija gena
Kalenik et al. (1997.) utvrdili su da dva prva cinkova prsta YY1 vežu YAF2. Razina YAF2 iRNK povećala se tokom in vitro diferencijacije ćelija skeleta i srčanog mišića pacova, za razliku od smanjene regulacije YY1. Prekomjerna ekspresija YAF2 u ćelijama mišjih mioblasta stimulirala je aktivnost miogenog promotora. Kotransfekcija i ekspresija YY1 u ovim ćelijama preokrenuli su stimulacijski učinak YAF2. YAF2 je također povećao proteolitsko cijepanje YY1 kalcija – aktiviranom proteazom m-kalpaina.
Kotransfekcijom i ekspresijom YAF2, GABPA i GABPB u ćelijama insekata, Sawa et al. (2002) utvrdili su da je YAF2 transkripcijski koaktivator za GABP.
