Cuscoko setioa edo Cuzcoko setioa, 1536ko maiatzaren 6an hasi eta 10 hilabetez luzatutako setio bat izan zen, honetan Manco Inka Yupanqui inka enperadorearen agindupeko inka gerlari gudarosteak Cusco hirian zeuden Hernando Pizarroren buruzagitzapeko 300 espainiar eta bertako inkak setiatu zituzten.
Emaitza berdinketa izan zen, alde batetik espainiar konkistatzaileak ez ziren setioa apurtzeko gai izan, eta bestetik, inkak ere ez ziren hiriaz jabetzeko gai izan.
